What is Tesamorelin?

Tesamorelin is a modified peptide analogue of growth hormone-releasing hormone (GHRH). It consist of the complete 44 amino acid sequence of GHRH, but modified by the addition of a trans-3-hexenoic group at the N-terminus. This modification increases the potency of the peptide in releasing growth hormone (GH) and also makes it more resistant to enzymatic degradation. Nonetheless, the peptide has a relatively short half-life of 26 – 38 minutes, although a single daily dose will produce sustained increase in GH. Tesamorelin is the only GH analogue in use that contains the complete 44 amino acid sequence of GHRH.

What does it do?

The main benefit is to reduce abdominal visceral (belly) fat. This is the type of fat that is most associated with risk of diabetes and cardiovascular disease. The peptide also decreases the amount of fat in the liver and in muscle. Fatty liver is also a major risk factor for chronic diseases, including serious liver diseases. Tesamorelin increases GH and IGF-1 levels which promote muscle gain while promoting fat loss. The peptide has been studied extensively for reduction of abdominal fat in HIV-infected patients. It is FDA approved for this purpose. The mechanism of abdominal fat loss is inhibition of the enzyme 11β-hydroxysteroid dehydrogenase type 1 which converts cortisone into the active steroid cortisol.

How to use it

Tesamorelin is administered by subcutaneous injection, usually in the abdomen. Rotate the site of injection to avoid local reactions. The usual dose is 1 – 2 mg given as single daily dose. Use five days a week for cycles of four weeks, with at least four weeks between cycles. Best given on an empty stomach before going to bed at night. Administer the injection at roughly the same time each day. Biov8 tesamorelin is supplied as 2mg/mL or 4 mg/mL in 5 mL vials. Two 4 mg/mL vials will provide a dose of 2mg (0.5mL) for a four week cycle. Tesamorelin can be used alongside repair peptides including BPC-157 and TB4. 

Is it safe?

Extensive studies in HIV-infected patients have shown that it is safe when administered for up to 52 weeks. Local reactions at the site of injection are seldom severe. Use with care in clients with a history of asthma or allergic reactions. Severe allergic reactions to the peptide have been reported, although these are rare. Other side effects are similar to those of other GH secretagogues. Arthralgia can affect up to 10% of users. This is usually mild and seldom limits treatment. Other systemic side effects include myalgia, fluid retention and paraesthesia. The peptide can increase appetite and should be used alongside an appropriate diet and exercise regimen. It can also increase blood glucose, although this is unlikely to be of major significance in short cycles of the peptide. Not indicated in women who are pregnant, planning a pregnancy or breastfeeding. Also contraindicated in those with active malignancy and use with care in those with a history of malignant disease. Tesamorelin can interfere with the metabolism of steroid drugs, ritonavir and cyclosporine. Take a full drug history from all clients. Use of the peptide is not permitted in professional sports competitors.

Potential Side Effects:

• Mild: Injection site redness, swelling, nausea, headache
• Serious (Rare): Increased blood sugar levels, carpal tunnel syndrome, hypersensitivity reactions

• Pregnancy or trying to conceive
• Breastfeeding
• Pituitary or hormone-related disorders
