Usage

1. Sample handling and requirements

1. Serum:
Whole blood samples collected in serum separation tubes were placed at room temperature 2 Hour or 4℃ Overnight, then 1000×g Centrifugation 20 Minutes, take the supernatant, or place the supernatant in -20℃ Or -80℃ Store, but repeated freezing and thawing should be avoided.

2. Plasma:
use EDTA Or heparin as an anticoagulant to collect specimens, and collect the specimens after collection 30 Within minutes 2-8℃ 1000×g Centrifugation 15 Minutes, take the supernatant to detect, or place the supernatant in -20℃ Or -80℃ Store, but repeated freezing and thawing should be avoided.

3. Tissue homogenate:
With pre-cooled PBS (0.01M, pH=7.4) The tissue is flushed to remove residual blood (lysed red blood cells in the homogenate can affect the measurement), and the tissue is weighed and cut into pieces. Combining the shredded tissue with the corresponding volume PBS (Generally according to 1:9 Weight to volume ratio, such as 1g The tissue samples correspond to 9mL Of PBS The specific volume can be appropriately adjusted according to the experimental needs and recorded. Recommended in PBS Add protease inhibitor) into a glass homogenizer and ground thoroughly on ice. For further lysis of tissue cells, the homogenate can be sonicated, or freeze-thawed repeatedly. Finally, the homogenate was mixed in 5000×g Centrifugation 5~10 Minutes,
Take the supernatant for detection.

4. Cell culture supernatant or other biological specimen:
Please 1000×g Centrifugation 20 Minutes, take the supernatant to detect, or place the supernatant in -20℃ Or -80℃ Store, but repeated freezing and thawing should be avoided.

Note: Hemolysis of the specimen will affect the final test result, so hemolyzed specimens should not be tested.

Two, Reagent Preparation

The kit should be removed from the refrigerated environment and equilibrated at room temperature before use.

20× Dilution of wash buffer: distilled water according to 1 : 20 Dilution, i.e. 1 Share 20× Wash buffer plus 19 Distilled water.

Three, Operation steps

1. Equilibration from room temperature 20min
Take out the required slats from the aluminum foil bag after that, and seal the remaining slats with ziplock bag and put them back 4℃ 。

2. Set up standard wells and sample wells, add different concentrations of standards to each standard well 50μL ;

3. The sample to be tested is added to the sample well 50μL ;
Blank holes are not added.

4. In addition to the blank wells, horseradish peroxidase ( HRP ) Labeled detection antibody
100μL Sealing the reaction hole with a sealing plate membrane, 37℃ Incubation in water bath or incubator 60min 。

5. Discard the liquid, pat dry on absorbent paper, and fill each well with wash liquid ( 350μL ), let stand 1min , throw off the washing liquid, pat dry on absorbent paper, and repeat washing the plate 5 Times (the plate can also be washed with a plate washing machine).

6. Substrate added per well A 、 B  Each 50μL , 37℃ Incubate in the dark 15min 。

7. Add stop solution to each well 50μL , 15min  Inside, in 450nm Wavelength measurement of each well OD  Value.

Calculation of experimental results

Based on the tested standard OD The value is the abscissa, and the concentration value of the standard product is the ordinate. Draw the standard curve on coordinate paper or with relevant software, and obtain the linear regression equation. The sample OD Values are substituted into Eq., and the concentration of the sample is calculated.

Species Reactivity Rat
Theory The kit uses a double-antibody one-step sandwich enzyme-linked immunosorbent assay (ELISA). To the coated microwells pre-coated with rat parathyroid hormone (PTH) capture antibody, specimens, standards, and HRP-labeled detection antibodies were sequentially added, incubated and thoroughly washed. The color is developed with the substrate TMB, which is converted to blue under the catalysis of peroxidase and to the final yellow under the action of acid. There was a positive correlation between the depth of color and rat parathyroid hormone (PTH) in the sample. The absorbance (OD value) was measured with a microplate reader at a wavelength of 450nm, and the sample concentration was calculated.

1. The standard concentrations are as follows: 2000 、 1000 、 500 、 250 、 125 、 62.5 pg/mL

2. After inspection of a large number of normal specimens, the normal concentration values of the specimens are all within the detection range provided by the kit, which are directly taken during the experiment 50μL Just load the sample. When some sample values exceed the maximum standard concentration, the sample can be appropriately diluted with sample diluent before conducting the experiment.
General Notes

1. Incubate in strict accordance with the specified time and temperature to ensure accurate results. All reagents must reach room temperature 20-25 °C prior to use. Store reagents in refrigeration immediately after use.

2. Incorrect plate washing can lead to inaccurate results. Make sure to drain the liquid from the wells as much as possible before adding the substrate. Do not allow the wells to dry out during incubation.

3. Eliminate the residual liquid and fingerprints at the bottom of the plate, otherwise it will affect the OD value.

4. The substrate color development solution should be colorless or very light in color, and the substrate solution that has turned blue cannot be used.

5. Avoid cross-contamination of reagents and specimens to avoid wrong results.

6. Avoid direct exposure to strong light during storage and incubation.

7. After equilibrating to room temperature, open the sealed bag to prevent water droplets from condensing on the cold slats.

8. Any reaction reagent cannot come into contact with the bleaching solvent or the strong gas emitted by the bleaching solvent. Any bleaching component will destroy the biological activity of the reaction reagents in the kit.

9. Expired products cannot be used.

10. If it is possible to spread diseases, all samples should be managed, and the samples and testing devices should be handled according to the prescribed procedures.

Storage Temp. Unopened kit, stored at 4 °C, shelf life 6 months.
Test Range 62.5 pg/mL-2000 pg/mL; Sensitivity: The lowest detection concentration is less than 10 pg/mL.

Color: Higher Level-Navy A
The design is adorable and I love the little bones and paw prints they put on the tabs. They are easy to slide and the colors are bright and admirable. There is different difficulty levels. The yellow circles are more on the easy side. The red ones are a bit more harder if you have a more intelligent dog. The three white tabs in the middle move in a 360 circle, there is one missing so you can slide the red tab in to hide the treats. I use my dog's kibble as treats; the kibble pellets are 1cm in length and 1cm in width. I can fit about 2 pellets in each tab hole, there are 14 small holes which you can hide the treats in total. Cleaning the puzzle It depends if you wash it by hand or put it in the dishwasher. I do both but I prefer the dishwasher because it is a little hard to clean by hand since there is so many nooks and crannies on the toy. Of course, it is dishwasher safe and I recommend putting it in by itself because you don't want to cross-contaminate the dog slobber to be on your dishes. I wouldn't put crushed or wet dog food in it because it could get in the sliding rails and jam it up or get moldy. If you are going to dry the toy, open all the tabs so they are exposed. If you cannot, that's okay. My dog's opinion I have two dogs, a 2-year-old chocolate Labrador named Maggie and a 7-year-old labradoodle named Rufio, Maggie takes about 1 or 2 minutes to finish the puzzle because she is super energetic. Rufio takes 3 or 4 minutes because he is calmer; but sometimes I have to help him just a little bit. It is suitable for all dog ages and I recommend it if your dog (s) are annoying you. They both love it and I give it to them for mental stimulation and as a reward. The bottom has "non-slip pads", but if I'm being honest they actually don't work that well. My dogs just slide it around.

Great design!
Color: Higher Level-Navy A
This Joansan Dog Puzzle Toy is exactly what I was looking for. My little MinPin is a worker and he likes having a job to do. I purchased this to entertain him when rolling his treat ball all over the place isn't practical. This disc has a non slip bottom and it's actually fairly heavy. It doesn't slide easily. So my little worker usually ends up picking it up and transporting it to different locations throughout the house. Obviously, individual results will vary, depending on size, personality, and breed of the dog using it, but it's been great for my lil guy. He gets a little physical and mental exercise. It's sturdy enough to withstand his scratching and chewing. My big dog likes it too. The size is perfect for food oriented cats too. My cat is too lazy to work for his food though. So he wasn't interested in it.

This feeder will surely make your dogs work for their food.
Color: Higher Level-Navy A
I recently tried this slow feeder (I purchased 2 feeders) for my two Corgis (25 & 30 lbs.). Overall, it’s a well-constructed product and definitely slows down eating as intended. The design encourages dogs to work for their food, which helps with digestion and keeps them engaged. That said, there are a couple of challenges I noticed. The kibble sometimes gets jammed in the sliding doors, which can make it harder for my pups to access the food. Also, the feeder doesn’t hold a large amount of kibble—I found myself needing to refill it 2–3 times per meal to accommodate both of my dogs. Finally, my pups had a tougher time reaching the kibble in the middle ring, which caused a bit of frustration. Despite these issues, it’s sturdy, safe, and does the job of slowing down fast eaters. I think it could work especially well for smaller dogs or those who eat less at mealtime.

He loves it, what else is there to say!
Color: Green
My dog loves this ball. Originally we used it as it is designed and put treats inside it to knock out and keep him busy, now he just loves to throw it around and fling it by the pieces of fabric that come out. He is generally a toy destroyer so the fact that it is still arounds shows that it is strong and was for sure worth the buy. Would 100% recommend for any dog that likes to keep busy!

Pupper shakes with excitement when I take this out
Color: Green
This is a very good toy for a super busy pup but take it away after they find the treats or it's getting destroyed (the fleece) totally worth the money and the five minute prep time for a play session
