Background

Peptide: N-glycosidase F (PNGase F) is an asparagine amidase produced by Flavobacterium meningosept-icum that serves as a useful tool in the research on protein N-glycosylation. Recombinant expression in E.coli. The cleavage site of PNGase F is the amide bond between N-acetylglucosamine (GlcNAc) and aspartate residues on the medial side of the glycoprotein, and converts aspartyl to aspartic acid on the enzymolysis protein. This product is often used for complete deglycosylation of antibodies and their associated proteins.

The PNGase F Glycan Cleavage Kit includes all components necessary to perform the enzymatic removal of almost all N-linked oligosaccharides from glycoproteins. The kit includes recombinant Peptide N-Glycosidase F(PNGase F) enzyme, which cleaves N-glycan chains at the innermost GlcNAc and asparagine residues of high mannose, hybrid, and complex oligosaccharides, and a 10X reaction buffer.

* One unit is defined as the amount of enzyme required to remove > 95% of the carbohydrate from 10 μg of denatured RNase B in 1 hour at 37°C in a total reaction volume of 10 μl.

Protocol

Denaturing Reaction Conditions:

1. Combine 1-20 µg of glycoprotein, 1 µl of Denaturing Buffer (10X) and H2O (if necessary) to make a 10 µl total reaction volume.

2. Denaturation is terminated by heating to 100 for 10-20min and cooling to room temperature.

3. Make a total reaction volume of 20 µl by adding 2 µl Reaction Buffer (10×), 2 µl 10% NP-40 and 6 µl H2O.

4. Add 1 µl PNGase F, mix gently.

5. Enzymatic digestion at 37 for 1h

6. Analyze by the method of SDS-PAGE

二. Non-Denaturing Reaction Conditions:

1. Combine 1-20 µg of glycoprotein, 2 µl of Reaction Buffer (10×) and H2O (if necessary) to make a 20 µl total reaction volume.

2. Add 2-5 µl PNGase F, mix gently.

3. Enzymatic digestion at 37°C for 4 - 24 hours.

4. Analyze by the method of SDS-PAGE

Guidelines

1.The target protein should be in a solution compatible with Rapid PNGase F activity. Avoid buffers containing SDS, as it inhibits PNGase F. Common stabilizing reagents such as Tween, Triton X-100, NP-40, octyl glucoside and non-detergent sulfobetaine, as well as traces of organic solvents, can prevent optimal rapid deglycosylation.

2.N-linked glycans containing core α1-3 Fucose are not cleaved by PNGase F.

3.For deglycosylation of native glycoprotiens, increased incubation time and increased amount of enzyme may be needed.

4.Optimize reaction conditions for each substrate.

5.If a larger amount of glycoprotein is used, scale up reaction volumes accordingly.

6.Although this product has been optimized for the rapid removal of N-glycans from antibodies, it can be utilized with various other glycoproteins.

Meeting my expectations....

I am a male with pretty large feet (Mens size 14US) and wanted some very warm slippers to wear around the house. I have nerve issues in my toes and if I don't keep my feet very warm they start to hurt. I wanted something both comfortable and warm. I received these a few days ago and so far they have done a great job at keeping my feet warm. The linings are thick and fluffy and the bottoms feel like they have a memory foam inner sole in them. The tops can either be worn up or rolled down. The actual soles are a soft leather-like material. They are thicker than normal cloth, but not hard like rubber soled slippers (I actually wanted something that was more of a slipper and less of a "shoe"). The only down side is that they don't really seem to have a left and right to them (both slippers seem identical), and for awhile no matter how I wore them, they felt a bit like they were on the wrong feet. However, they seem to have formed themselves to my feet and now (through wearing them) it definitely feels like there is a left and a right. I don't know how long these will last, but they were not expensive, so if I get a few months of wear from them, I will be happy and will just order another pair when these wear out. They aren't the most attractive slippers in the world, but they aren't horrible looking and they are very functional.

for polar fleece slippers with leather soles, these are quite nice

I bought these to replace some Mug Lugs slippers last fall after they had started to come apart from barely a year of use (they were knitted) and have been wearing them constantly since then. The polar fleece does easily pill somewhat but get a hard use and are quite comfortable too. I wear these in the house most of the time as my feet do not like to wear shoes all day. They are comfortable, and do seem to hold up well. The only other issue is until I get my office chair mat replaced, these catch and can cause problems and have a little, but not enough to worry too much about. They supposedly have an anti slip design on the bottom, but if flour falls on the kitchen floor, not so slip resistant. I chose these as one, they are red, but also plaid and like them a lot. Hopefully, I get more than a years use out of them. I have had several polar fleece sock type slippers similar to these in the past, and the last polar fleece pair in Dk Blue and from Land End finally after several years of constant use, the stitching began to come apart and the sole was separating from the body of the slipper on one slipper. They do keep the feet warm, with or without socks and can, at times, get my feet too warm but I enjoy them quite a bit.
